Kelembak (Rheum officinale Baill), known as rhubarb, is herb that the roots are often used as a basic ingredient of traditional medicine. The herb roots contain rhein (C15H8O6) as a marker compound which is included in a group of anthraquinone. In this research, the production of marker compound was carried out from the dry roots powder of rhubarb. The roots extraction was done through maceration using ethanol 80%, while the separation was performed by column chromatography. The isolate compound produced was yellow powder formed. After characterization by UV-Vis, FT-IR, 1H-NMR and13C-NMR, it could be concluded that the isolate compound was Rhein (C15H8O6). Production of rhein marker compound from rhubarb roots dry powder was gained 0.11% yield.
